RESUMO

Objective:To explore the application effect of case-based learning (CBL), teaching mode combined with 3D printing in clinical teaching of sacral tumors.Methods:A total of 108 undergraduate interns and standardized residency training students who studied in our hospital from 2017 to 2018 were divided into the CBL teaching group ( n = 53) and the CBL combined with 3D printing teaching group ( n = 55) according to their study time. The combined teaching group used computer tomography (CT) data to reconstruct and print out a 3D model of sacral tumors based on CBL, and performed preoperative teaching on the invasion of the surrounding tissues of the tumor. The scores of the students in the two groups were evaluated respectively, and the students were surveyed by self-identification questionnaire (learning interest, self-learning ability, teamwork ability, comprehensive analysis ability and clinical thinking ability). The t-test (one-sided) was used for comparison between groups using stata 14.0. Results:The score of CBL teaching group (75.90±6.70) was lower than that of CBL combined with 3D printing teaching group (83.60±7.40). In terms of critical thinking ability evaluation, self-learning ability, learning interest, comprehensive analysis ability and clinical thinking ability, the CBL combined 3D printing teaching group was superior to the CBL teaching group, and the difference was statistically significant ( P<0.001). In terms of teamwork ability, there was no statistical difference between the two groups. Conclusion:The CBL teaching mode combined with 3D printing can improve academic performance, students' learning interest and clinical thinking ability of sacral tumors in the teaching of undergraduate interns and standardized residency training students.

Objective To evaluate the effect of TanshinonneⅡA preconditioning on nuclear factor kappa B ( NF-κB ) signaling pathway in N9 cells subjected to oxygen-glucose deprivation and restoration ( OGD∕R) . Methods N9 cells cultured in vitro were divided into 4 groups ( n=16 each) by using a ran-dom number table method: control group ( C group) , OGD∕R group, vehicle group ( V group) and Tanshi-none ⅡA preconditioning group ( TP group) . N9 cells were subjected to O2-glucose deprivation for 4 h fol-lowed by restoration of O2-glucose supply for 24 h to establish the OGD∕R model. Group C receive no treat-ment. In group TP, N9 cells were cultured in medium containing Tanshinone ⅡA ( final concentration 5 mg∕L) for 24 h before the model was established. Dimethyl sulfoxide was added to the culture medium in group V. At the 24 h of reoxygenation, the level of lactate dehydrogenase ( LDH) and nitric oxide ( NO) in culture medium was detected by spectrophotometry, cell apoptosis was assessed by using TUNEL, and the expression of NF-κB in nucleus was detected by Western blot. The apoptosis rate was calculated. Results Compared with group C, the levels of LDH, IL-β, TNF-α and NO in culture medium and apoptosis rate were significantly increased, and the expression of NF-κB in nucleus was up-regulated in OGD∕R, V and TP groups ( P<0. 05) . Compared with group OGD∕R, the levels of LDH, IL-β, TNF-αand NO in culture me-dium and apoptosis rate were significantly decreased, and the expression of NF-κB in nucleus was down-reg-ulated in group TP ( P<0. 05) , and no significant change was found in the parameters mentioned above in group V ( P>0. 05 ) . Conclusion The mechanism by which Tanshinonne ⅡA preconditioning reduces OGD∕R injury to N9 cells is related to inhibiting NF-κB signaling pathway.

RESUMO

Objective To evaluate the effect of Xingshi decoction combined with warming needle moxibustion combined with conventional western medicine on acute knee arthritis.Methods A total of 86 patients with acute knee arthritis were randomly divided into two groups,43 in each group.The control group took diclofenac capsules orally and methotrexate once a week,while the observation group took Xingshi decoction and wanning needle moxibustion on the basis of the control group.Both groups were treated for 2 weeks.The Rheumatoid factor (RF),high sensitive C reactive protein (hs-CRP) and IL-1β were detected by ELISA,and pain rating index (PRI),VAS and present pain intensity (PPI) were evaluated by McGill pain questionnaire (SF-MPQ).The knee function was assessed by the evaluation scale,and the curative effect was evaluated by 28 joint disease activity-erythrocyte sedimentation rate (ESR) scores.Results The total effective rate was 88.4% (38/43) in the observation group and 62.8% (27/43) in the control group.There was significant difference between the two groups (Z=2.678,P=0.007).After treatment,ESR (30.95 ± 3.14 mm/h vs.55.13 ± 3.28 mm/h,t=34.919),RF (81.36 ± 5.13 U/ml vs.121.73 ± 7.25 U/ml,t=29.807),hs-CRP (10.13 ± 3.12 mg/L vs.34.26 ± 3.17 mg/L,t=35.575),IL-1β (58.13 ± 3.12 pg/ml vs.96.76 ± 3.17 pg/ml,t=56.952) of the observation group were significantly lower than those of control group (P<0.01);and PRI,VAS and PPI scores of SF-MPQ in observation group were significantly lower than those of control group (t values were 19.661,11.056 and 23.524,respectively,P<0.01) and the scores of walking,swelling,sense of stability,strangulation and stair-climbing were significantly higher than those of the control group (t values were 11.071,13.464,9.102,9.804,10.012,respectively,P<0.01).Conclusions The Xingshi decoction combined with warming needle moxibustion is effective in treating acute knee arthritis.It can effectively relieve pain and inflammation of patients,thus improving knee joint function.It has certain clinical application value.

OBJECTIVE:To observe the effects and safety of Congrong yishen granule combined with Donepezil hydrochloride tablet on mental state and quality of life in patients with Parkinson disease with dementia(PDD). METHODS:A total of 60 PDD patients were randomized into control group and treatment group,with 30 cases in each group. Both groups were given drugs for Parkinson disease. Control group was additionally given Donepezil hydrochloride tablet 5 mg,qd;treatment group was additionally given Congrong yishen granules 2 g,bid,on the basis of control group. The treatment lasted for 6 months in both groups. MMSE, MoCA,ADAS-Cog,ADL-R and TCM symptom score were observed in 2 groups before and after treatment. The occurrence of ADR were compared between 2 groups. RESULTS:Before treatment,there was no statistical significance in MMSE,MoCA, ADAS-Cog,ADL-R or TCM symptom score between 2 groups (P>0.05). After treatment,MMSE and MoCA score of 2 groups were increased significantly,while ADAS-Cog,ADL-R and TCM symptom score were decreased significantly;the treatment group were better than the control group,with statistical significance (P<0.05). No obvious ADR was found in 2 groups. CONCLU-SIONS:Congrong yishen granule combined with donepezil hydrochloride tablet can significantly improve mental state and cogni-tive ability of PDD patients,and relieve clinical symptoms with good safety.

RESUMO

Objective To investigate the clinical efficacy of auricular point therapy for chronic fatigue syndrome.Methods Eighty patients with chronic fatigue syndrome were randomly allocated to treatment and control groups, 40 cases each. The treatment group received ear acupuncture plus auricular plaster therapy and the control group, conventional acupuncture. The overall symptom was scored, the Fatigue Assessment Instrument (FAI) score was recorded and immunoglobulins (IgA, IgG and IgM) were measured in the two groups before treatment and after four courses.Results There were statistically significant pre-/post-treatment differences in the overall symptom score, the FAI score and immunoglobulins in the two groups (P<0.05). There were statistically significant post-treatment differences in the FAI score, and IgA and IgG between the treatment and control groups (P<0.05). Conclusion Auricular point therapy is an effective way to treat chronic fatigue syndrome.

Background and purpose:Tumor metastasis is a main reason of breast cancer patients’ death. This study aimed to discuss whether or how the transcription factor ETS2 regulate CXCR4 transcription and the molecular mechanism of ETS2 modulating transcriptional activity of CXCR4 gene in human breast cancer cells. Methods:In MCF-7 breast cancer cell lines and MDA-MB-231 breast cancer cell lines, through transient transfection, as well as RNAi technology, the expression of ETS2 was overexpressed or inhibited was detected. RT-PCR and ELISA was used respectively to detect CXCR4 mRNA expression and protein level. Luciferase reporter gene assay was applied to detect CXCR4 promoter activity, and ChIP for detecting the amount of ETS2 protein binding to CXCR4 promoter. Two binding sites of CXCR4 promoter were mutated to detect the impact on the activity of CXCR4 promoter by gene mutations. Results:After transfected with ETS2 expression vector in MCF-7 and MDA-MB-231 breast cancer cell lines, the mRNA expression and protein level of CXCR4 were elevated. The result of luciferase reporter gene assay indicated that overexpression of ETS2 activated CXCR4 promoter. ChIP assay demonstrated that the amount of ETS2 protein binding to CXCR4 promoter increased after ETS2 transfection. This result indicated that ETS2 may activate CXCR4 promoter through directly binding with CXCR4 promoter. Inhibition of ETS2 expression using RNAi could significantly attenuate CXCR4 promoter activity and reduce expression of CXCR4. Two ETS binding sites of CXCR4 promoter were mutated and the result of luciferase reporter gene assay proved that, an arbitrary point mutations attenuated CXCR4 promoter activity, while mutation of both binding sites further attenuated CXCR4 activity. Conclusion:In MCF-7 and MDA-MB-231 breast cancer cell lines, overexpression of ETS2 could activate CXCR4 promoter and the transcription of CXCR4 through directly binding to two ETS2 binding sites (-540 to-535 and-240 to-235) of CXCR4 promoter.

RESUMO

Objective To evaluate the expression and function of mammalian target of rapamycin (mTOR) and its substrate kinase P70S6K in mTOR/P70S6K signal pathway in cervical carcinogenesis. Methods The expression of mTOR and P70S6K in normal cervix uteri or cervical cancer was detected by the immunohistochemistry and RT-PCR methods. Results Compared to those in normal cervix uteri, both mRNA and protein of mTOR/P70S6K in cervical carcinomas were significantly increased (P<0.05). There was a positive correlation between mTOR gene and P70S6K gene expression in cervical cancer (r=0.746, P<0.001). Conclusion High expression of mTOR and P7OS6K may play an important role in the pathogenesis and development of cervical cancer.
